The overall lead raced past 450 as Watson launched a morning onslaught against the English bowlers.
Watson was 29 not out at the start of play but took just 40 balls to advance to his fourth Test century, taking 22 runs off one Graeme Swann over.
His innings came to an end in farcical circumstances when he was run out for 103, from 108 balls with 11 fours and five sixes.
Watson lofted a Tim Bresnan delivery straight up in the air and Ian Bell dropped the catch, but the bowler picked up the ball and threw the stumps down, with the Australian well out of his ground having assumed he would be caught.
