"It's going to be a huge moment for Hashim to play in his 50th Test, but I know it will not unnerve him," Smith told the Afrikaans daily Beeld here today.
"His discipline and ability to stay calm under pressure is exactly what makes him such a fantastic player," said Smith.
Smith also added that Amla was by far the best batsman in the world in 2010 in all forms of the game.
"I believe (Amla) will end his dream year with a bang. To have him in the team is an incredible privilege because he was, especially in 2010, at the forefront of our success. He established himself as one of the leading players in the world," the captain said.
Amla scored 1,200 Test runs in 2010, while hit 1,058 in the one-day format.
Smith confirmed that South Africa were full of confidence after defeating India as the top team in the world with an innings and 25 runs at Centurion last week in the first Test.
"It's such a big advantage to win the first Test so convincingly, but India will come back strongly," said Smith.
