Monfils, the world number 17, has not played since the end of May and missed both the French Open and Wimbledon.
He will be replaced in the French team for the Olympic tennis event, which takes place at Wimbledon from July 28 until August 5, by Julien Benneteau, the International Tennis Federation (ITF) said.
Jo-Wilfried Tsonga, Gilles Simon, Richard Gasquet and Benneteau will play singles at the Games with Tsonga teaming with Michael Llodra and Gasquet with Benneteau in the doubles.
Monfils's withdrawal from the Games comes just a day after world number three Rafael Nadal also pulled out.
