The ninth seeds stunned veteran top seeds Mike and Bob Bryan 5-7, 6-4, 7-6(9), 7-6(5) in two hours and 35 minutes to book a place in the last four of the tournament on Tuesday.
"Still have adrenaline running, feels great to be through to the semis. Thank you all for the support," Bopanna tweeted. (Roger Federer reaches 10th Wimbledon semis)
Next up, Bopanna and Mergea will take on fourth seeds Jean-Julien Rojer of the Netherlands and Horia Tecau of Romania for a place in the final.
